The EEG neurobiomarker platform from Beacon Biosignals is designed to find and scale novel therapeutic paradigms for patients with neurological and psychiatric disorders. Beacon Biosignals identifies neurological indicators of pharmacological action and efficacy using a world-class clinical Electroencephalogram (EEG) database, unique artificial intelligence-based algorithms, and analytical capabilities. Different EEG profiles are linked to neurodevelopmental status and various neurological diseases, and neurologically active treatments can alter them. To help overcome the challenges of transferring PK/PD models from preclinical animal studies, the platform quickly assesses these effects and informs dose selection in humans.

Cyclerion and Beacon Biosignals previously collaborated to analyze data from a Cyclerion clinical translational pharmacology trial of CY6463, which showed clear impacts on EEG parameters linked to aging and disease. These findings were recently presented at the American Academy of Neurology’s annual meeting (Glasser et al., 2021). The inclusion of the Beacon Biosignals technique in ongoing and planned Cyclerion patient trials is expected to help the company’s primary clinical program, CY6463, as well as CY3018, a next-generation preclinical study.
CY6463 is a Central Nervous System (CNS)-penetrant, oral, first-in-class sGC stimulator being developed for neurological illnesses linked with cognitive impairment. CY6463 was created to target various pathophysiological characteristics linked with cognitive impairment in neurodegenerative and neuropsychiatric illnesses. Initial CY6463 clinical tests showed good safety and tolerability and pharmacologically appropriate drug exposure in the cerebral spinal fluid. Furthermore, CY6463 showed encouraging effects on EEG, neuroinflammation, and other neurophysiological markers, all of which support further clinical development.
Patient studies in Alzheimer’s disease with vascular pathology (ADv) and Mitochondrial Encephalomyopathy, Lactic Acidosis, and Stroke-like events (MELAS) are now underway, with a study in patients with Cognitive Impairment Associated with Schizophrenia (CIAS) anticipated to begin soon. EEG and other neurological biomarkers will be analyzed extensively in these studies.
