Pathios Therapeutics reveals that it had been awarded £350K in the form of a Smart Grant from Innovate UK to boost its cancer immunotherapy program.
FREMONT, CA: Pathios Therapeutics Limited, an innovative biotech company prioritizing the production of first-in-class cancer therapies, has revealed that it had been awarded £350K (approximately $475K) in the form of a Smart Grant from Innovate UK, the UK government's innovation agency, to boost its cancer immunotherapy program targeting the innate immune checkpoint, GPR65. Pathios will coordinate with researchers from the University of Oxford's Department of Oncology for this project to develop the primary tools necessary to accelerate the development of small-molecule GPR65 inhibitors for treatment-resistant melanoma.

The introduction of immunotherapy agents that target T-cell checkpoints (PD-1/CTLA-4) has brought important improvement in the long-term survival of several melanoma patients. However, a limited percentage of patients received sustained benefits from these treatments, and it has become a continuous challenge to discover more therapies for the other non-responsive patients.
Recent groundbreaking science indicates that the disarming of innate immune cells called tumor-associated macrophages (TAMs) by the acidic microenvironment found in advanced tumors is a fundamental explanation why certain melanoma patients do not react well to anti-PD-1 therapies. When the pH-sensing receptor GPR65 on TAMs is activated by acidic pH, it suppresses several pro-inflammatory genes, causing these cells to transform from immune-stimulating to immunosuppressive. When analyzed in The Cancer Genome Atlas (TCGA), a small proportion of the population with inactivating polymorphisms showed stratified association with survival, highlighting the role of the GPR65 pathway in cancer. The Pathios Macrophage Conditioning strategy intends to use small-molecule GPR65 inhibitors to reverse pH-dependent immunosuppressive signaling in most of the patients who do not have this genetic alteration.
